In vitro study | Aptopia Activator 2 (20 μm) was at a reduced cyto c concentration, increasing the Apaf-1 fraction in apoptotic bodies 1.5-fold to 33%. Apoptosis Activator 2 (20 μm) was at a reduced level of cyto c, increasing the degree of activation of caspase-3 4-fold. Apoptosis Activator 2 strongly induced caspase-3 activation, PARP cleavage, and DNA division, and eventually killed cells with an IC50 of 4 μm. Apoptosis Activator 2 induces Apoptosis in PBL, HUVEC, Jurkat, Molt-4, CCRF-CEM, BT-549, MDA-MB-468 and NCI-H23, with IC50 of 50 μm, 43 μm and 4 μm, respectively, 6 μm, 9 μm, 20 μm, 44 M and 35 M. Apoptosis Activator 2 inhibited most of the tumor cell lines tested, with 50-100% inhibition of cell growth at 10 μm. Apoptosis Activator 2 induces cell death by triggering the formation of apoptotic bodies. The expression level of En1 had no significant effect on the survival rate of the ventral midbrain in culture. Apoptosis activator 2 (10 μm) induced Apoptosis in AGS cells was evaluated by DNA ladder electrophoresis and TUNEL assay for Apoptosis. Apoptosis activator 2(10 μm) enhanced the induction of Apoptosis by anti-TROP2 antibody conjugated liposomes. Cyclohexamide (10 μg/mL) or zVAD (50 μm) acts on cultured neurons to significantly prevent the toxicity of Apoptosis Activator 2. Apoptosis activator 2(3 μm) causes many neurons to undergo nuclear condensation, suggesting that cell death involves Apoptosis. DHT(10 nM) or E2(10 nM) acts on cultured neurons to significantly prevent the toxicity of apotosis Activator 2. |
